Gamble, drafted 28th overall in 2004 out of Ohio State, immediately became a starter for the Panthers. In his rookie season, he recorded 75 tackles, six interceptions, 14 passes defended, a forced fumble, and a fumble recovery. The Panthers front office recognized his value, awarding him a six-year, $53.5 million contract extension in November of that year.

The 2010 season was challenging for Gamble due to a high ankle sprain, causing him to miss several games and marking the first time in his career he went without an interception. He rebounded in 2011, leading the NFL in target rate, with opposing quarterbacks testing him on only 9.1% of drop backs.
